It's practically the same. The both mean "location" or "point where someone is". Plek is just more informal than plaats. You can ask a friend "een plek vrij te houden" (literally to save a spot), but when you ask someone you don't know too well, you'd rather ask "een plaats vrij te houden". It's the same, but you use "plek" in a more informal way.
