It can be physical work but also can be work that involves thinking, learning, writing etc.

Pareciera que "remangarse las mangas" fuera un pleonasmo ya que también puedes remangarte los pantalones :) Estoy de acuerdo con Antonio, "manos a la obra" encaja bien. Igualmente puedes decir: A trabajar / Hora de trabajar / Pongámonos a trabajar
