Yes. It is slightly formal, and you hear it mostly in written English, but it is also used in conversational English.
"He has a good command of Spanish." = He knows Spanish very well.

I would say it's a pretty common usage if we speak about a language mastering.
Ex: " She has a good command of that language ".
