No, there is no difference at all. The pronunciations are identical. You can check in a print or online dictionary. Similarly, there is no difference between "knot" and "not," or "knight" and "night." The explanation is that English spellings sometime preserve the way words were pronounced centuries ago.

Not at all. They're the same. "Knew" has a silent K, but both are pronounced as /nju:/.
