What is the difference between these two phrases? When should I use each one?

They have the same meaning, you can use both, it is just a different way to ask.
note:
Che ora è ? Sono le 7 or e` l'una.
Che ore sono ? Sono le 7 or e`l'una.
The answers are the same.
