Grammatically, the difference is that in the first phrase the verb has no direct object whereas in the second phrase, "us" is the direct object. "Let go" means "allow to go", or to release. As a command, "Let go!", it means "Release!". With a subject such as "He let go" it means "he released". "Let us go" means the same but specifies that the thing released is "us". Spoken to someone else (who is not part of "us") it means "allow us to depart". Spoken among ourselves, it means "Let us get going!".
