Can we say "Who are happy?"

Not on its own. If you were asking a question, you'd have to say "Who is happy?".
"Who are happy" can only be correct when it's part of a bigger sentence e.g. "People who are happy are..."

If that was the entire question, it would be ' Who is happy ' ? ( if you were asking a group, or said in a classroom )
