Eliminate "out of six people" from the sentence. It will be easier for you to see the correct verb required. Take a look at below examples .
One out of six people is sad. One is sad.
Two out of six people are sad. Two are sad.
Six out of six are sad. Six are sad.

"One out of six people is..." It's 'is' and not 'are' because you're talking about that one person, not all 6 of them. So it has to be singular. However, if you were two say "two out of six people...", then you would say "two out of six people are..." because it is no longer singular. Does that make sense?
