What's the meaning of "day-off "? How to use it? Is "3 days-off" right?

Day Off is a day you are not required to work. Can be a Saturday or Sunday generally... but sometimes you can take a day off during the middle of the week and have a "day off" then. Yes, you can say 3 days off but dont use "-"

A day-off is when you don't have to work, a day off work. "Is it your day off tomorrow?".
