"To tamp" means to pack down, press down or flatten down a material. You'd normally hear this verb used in smoking tobacco, manufacturing certain products or making espresso coffee.

It means that he was pressing the cigarette against his knuckle so that the tobacco would be more tightly packed inside the cigarette.
