Are both sentences OK?
I can't remove this splinter from my foot.
I hava a splinter on my foot, and it won't come out.

Yes, they both sound ok. You'd need to say 'in my foot' and not 'on my foot' in the second sentence though: I have a splinter in my foot and it won't come out.
