Est-ce que tu peux me le donner?
Tu peux me le donner s'il te plaît?
Those all mean Can you give it to me (please)?
The option with m'en donner is for the idea of "some"...as in some coffee, some cheese, some soup, etc.
- Il y a encore du café si quelqu'un en veut. There's still some coffee if anyone wants some.
- Oh très bien. Peux-tu m'en donner? Oh very good. Can you give me some?
