Are they both correct? 'Who knows most?' and 'Who knows the most?'

Only “Who knows the most?” is correct.
There is a difference in meaning between “most” and “the most”.
“Jack knows most of the people in this room” (the majority of the people)
“Jack is the person I know who knows the most about the people in this room.” (he has greater knowledge)
