I've to respond to email and i want to say if this expression "sure, i can do it" is an expression informal?

Yes, it is informal.
Since the second sentence ( "I look forward to receiving the cover letter.") is formal, you should say something like "I will be happy to take care of it.", "I will gladly take care of it.", etc.
