You can use both, it depends on the context. 'From' would be used in the context of using something that has some connection to the outside world.... 'work online from the comfort of your own home' but 'surf the internet in the comfort of your own home'. Basically 'from' implies some relationship with the outside world, and 'in' doesn't, although there are probably some exceptions.

Hi Megumi, There are two different prepositions here, and there is a nuance in their usage. "in" is often used to indicate location, while "from" is commonly used to indicate a separation of distance or a point of origin. Therefore, imagine that you are shopping online while at home. "Now you can enjoy the benefits and convenience of online shopping in the comfort of your home. (Focus is on the fact that you are in your comfortable house.) Now, imagine that you have a home office and you have the flexibility of working from home (away from your company office) on certain days of the week. "Now you can enjoy the benefits and flexibility of working from the comfort of your home. (Focus is on the fact that you are away from your company.) Both make sense. I would argue that you would use 'from' if it is something that you would ordinarily have to travel for and for something more static. Nut, really, both are fine.

Either makes sense, i think the 'correct' phrase is from the comfort of your own home (or at least the one I personally have heard more often), but in reality you would get native speakers saying it both ways, i'm sure.
