What is the difference between pour and spill?

'Pour' is usually deliberate; 'spill' is an accident or an overflow.
For example: "Can you pour the coffee into those cups, please? But be careful! Don't spill any coffee onto the table".

You pour something intentionally. You spill something unintentionally.
"I poured milk into the glass."
"I spilled milk all over the floor."
