What des it mean? They have blood in their eyes. Does it mean, They are so mad to kill someone.

If literal, then it means there is blood (actual blood) in their eyes. If figurative, then yes, it means they are very angry. You may more commonly hear the saying They are seeing red to indicate anger.

Probably, but it depends on the context. Their eyes are red, burning with anger, ready to kill.
