A "trying time" tests you.  It's a challenge.

A "tiring time" exhausts you.

"Through and through" means thoroughly, all the way through:  "I'm tired through and through".  This has a sense of poetry about it, and I remember this used in Lewis Carroll's poem "Jabberwocky".
