"Cup runneth over" is a reference to a Bible verse (somewhere in Psalms, I believe, but I'm too tired to look it up right now). It basically means having more than one could possibly need (i.e. God is pouring more wine than can fit into a cup). As a whole, this would mean' "I assure you, I am lucky/ blessed and do not lack anything I need."

"My Cup Runneth Over" is a quote from the Hebrew Bible[1] (Psalms 23:5) and means "I have more than enough for my needs," though interpretations and usage may vary. Notably, it can be employed sarcastically to indicate that someone, e.g. one's host, is being less than generous.
