Do you mind doing this? = The person you are speaking will do this (if they don't mind).
Do you mind ME (or MY) doing this? = You will do this (if the person you are speaking to doesn't mind).

The first one is saying that you (the person speaking) are going to do it. The second one is asking the other person to do it.
