Mizu, kudasai. or Mizu kudasai.
Both are OK. We often leave the particle wo out, as you said. If you use wo and say "Mizu wo kudasai.", this is formal and more correct.
Here adding the punctuation ten helps emphasize "water". Hope this helps.
