1) He asked me to help him.
2)He requested me to help him.

#2 is WRONG. You must use "asked".

When you ask someone to do something you can use 'request' instead of 'ask' but it is more formal. However, there is no difference in meaning
Although both sentences are correct the one with 'request' could also be written 'He requested help from me.'
