Really nothing. Just avoid confusing the phrases (ie. never write "despite of"... completely wrong).

you can put a sentence following "despite", but only words can follow "in spite of". for example: Despite the rain was heavy, I still went to school", " In spite of the heavy rain, I still went to school." Hope it will help you.
