There are two types of negation for a verb in Korean.
1. 안 + verb : do not (verb)
2. 못 + verb : cannot (verb)

For example, 
나는 복숭아를 안 먹는다. I don't eat peach.(I don't want to eat peach. ) - for the will of subject - ACTIVE
나는 복숭아를 못 먹는다. I cannot eat peach.(I want to eat peach, but I am unable eat peach for several reasons) - for situation - PASSIVE

Each from has long form
1. 안 + verb = verb + 지 않다 
2. 못 + verb = verb + 지 못하다

For example,
나는 복숭아를 안 먹는다. = 나는 복숭아를 먹지 않는다.
나는 복숭아를 못 먹는다. = 나는 복숭아를 먹지 못한다.

Same rule applies to the verb 이해하다(to understand)

저는 그것을 안 이해해요 = 저는 그것을 이해 안해요. = 저는 그것을 이해하지 않아요.
it literally means "I don't understand it.". It has nuance "I don't want to understand it."

저는 그것을 못 이해해요. = 저는 그것을 이해 못해요. = 저는 그것을 이해하지 못해요. 
it literally means "I can't understand it.". It has nuance "I want to understand it. but I'm unable to understand it".

As I know, English word "understand" is not used with "cannot" but used with "don't".
But, don't stick to the English word "understand".
All Korean verbs can be joined with 안 and 못.

When negating a verb, you should choose between 안 and 못 depending on the will of the subject.

@Jason. Lee But I don't get why 이해하지 않다 is unnatural? Wouldn't it be (in your words) "denying a previous action?" Is there no explanation to this? If so, how can I know when I need to use 못?

1. 어떤 행동을 안하다. (Do not do something)
만약 참석하지 않을거라면 미리 알려 주세요. (If you are not going to attend, please let me know in advance)
공부도 않고 뭘하고 있는거야? (What are you doing without studying?)

2. 앞의 행동을 부정함. 동사 + ~지 않다. (Denial of previous actions)
난 저녁을 먹지 않습니다. (I not eat dinner)
난 일기를 쓰지 않아요. (I do not write a diary)
난 거기에 가지 않을꺼야. (l  not go there)

3. 앞의 상태를 부정함. 형용사 + ~지 않다. (Denial of previous states)
공부는 쉽지 않다. (Studying is not easy)
나는 예쁘지 않아. (I'm not pretty)
그것은 좋지 않다. (It is not good)
