te lo dire en español la prime i do have a couple of questions though significa aunque yo si tengo par de preguntas ese do es como para afirmar y though tiene un significa como de pero (but) asi que puedes decir but i have a couple questions y no poner el though al final pero en muchos casos va al final de la oracion como tu pregunta.

"I have a couple of questions, though"
Both are correct when you add the comma before 'though'.
'Do' is added for emphasis. It would be stressed slightly in the sentence.
