what does "take the second left and the third right" mean?
Follow the main road, take the second left and the third right.
take the second left=take the second turning on the left?

Yes, don't turn left the first time there is a left turn. Turn left the second time.
There should be a "then" added to make it more clear in my opinion. As you can't possible take the second left and the third right on the same road.
"Follow the main road, take the second left and then the third right."
