What is the difference between saying...
"los llamo" and "llamo sus" (I'm trying to say "I'm calling them") ???

You want, "Los llamo." or "Estoy llamándolos."
"Sus" is an adjective in the plural meaning, "his/her/its/your (formal)/their", so "llamo sus" by itself means nothing grammatical.
