What is the difference between the two words? From my understanding they both mean beside.

となり basically means something or someone neighboring. おとなりさん means your neighbors.
よこ means a side or horizontal direction. For example, よこになる means placing yourself horizontally, namely to lie down.
となりのひと and よこのひと both mean the same thing.
