Can we use "on the other hand" as an alternative to "however" without using "on the one hand"?

Yes, it's perfectly acceptable and intelligible, although somewhat informal (a dictionary might encourage you to use both, for instance). In fact, it can sometimes sound stilted to use both "on the one hand" and "on the other hand."
