Грязнуле: Actually, in America, we do not say "a scruff" at all. If you said that, people would not understand. You can say "a pig" or "a slob" if you want to be harsh. (Note: these two words will offend people.) If you want to be mild, you can say the person is "messy".
Чистюле: You can certainly say "clean freak" or "neat freak" - this is correct slang.

Если вы хотите получать ответы от нейтивов, надо соответственно ставить вопросы. Например, how do you call a person who can't stand a mess in the room and is always ready to do some housework?
Самый близкий перевод к грязнуле будет a scruff, к чистюле a tidy person. Нашего "уменьшительного" оттенка в этих словах нет, конечно. Если хотите к чистюле добавить пренебрежительный оттенок, скажите neatnik, neat freak.
