Which the question is OK? Which is correct? 'Who knows the answer?' or 'who does know the answer'?

The first one is correct. Who knows the answer.

"Who knows the answer" is correct. We usually don't use a "do" form with a verb in a positive statement, only in a negative one (like I'm doing here!).
