You can say "il n'y a personne qui parle français dans le bureau" (there is no one who speaks French in the office) or "personne ne parle français dans le bureau" (no one speaks French in the office). Hope this helps! :)

La phrase la plus courante serait : "S'il vous plaît ! Est-ce qu'il y a quelqu'un qui parle français dans ce bureau ?" Better than yours I think. "Est-ce qu'il y a quelqu'un" is better than "Il n'y a personne". Seems more polite. La négation n'est pas bienvenue dans cette phrase.
