It means " at that particular moment in time". 
"As of five minutes ago, your time was up." = "At the instant in time that passed five minutes ago, your time was up."
"As of now, I can go to the store with you." = "At the current moment in time, I can go to the store with you."
" As of now, I can go to your party next month." = "Right now I believe I can go to your party."
Please note that when this term is applied to events that take place in the future, such as the last example above, the person may really be saying, "Based on what I know right now, I can do that, but things might change.".
