Is this sentence correct and natural?
Ecotourism has great potential to widen participation by different age groups in it.
(I'm not sure about the preposition "by" and the word order.)

Your sentence is really good! Any English speaker would understand you perfectly. To make it slightly more natural, I think I would say:
Ecotourism has great potential to INCREASE THE participation OF different age groups.
