There are several ways to say this. "I am separated from my wife" or "my wife and I are separated", "I got separated from my wife" and "I separated from my wife" are also possible.

Susan is right, there are several correct ways to say it. However "separate my wife" is incorrect; that sounds like you're going to cut her into pieces, haha!
